Produced primarily by <strong>B-lymphocytes<\/strong>, antibodies have the ability to recognise and neutralise antigens, which are foreign substances in the body, such as <strong>bacteria, viruses and toxins.<\/strong><\/p>\n<p><strong>Structure and Function of Antibodies<\/strong><\/p>\n<p>Antibodies can exist in two main forms: membrane-bound or secreted. <strong>Membrane-bound<\/strong> antibodies act as receptors that initiate the activation of <strong>B-lymphocytes<\/strong> upon binding to an antigen. On the other hand, secreted antibodies play a key role in<a href=\"https:\/\/immunostep.com\/immunology\/\"><strong> humoral immunity <\/strong><\/a>by activating various effector mechanisms to eliminate bound <strong>antigens.<\/strong><\/p>\n<h3><strong>Antibody diversity<\/strong><\/h3>\n<p>One of the most <strong>salient features<\/strong> of antibodies is their great diversity. The <strong>antigen-binding<\/strong> regions on antibody molecules are highly <strong>variable.<\/strong> This allows an individual to be able to generate millions of different antibodies, each with specificity for a particular antigen. This diversity is essential for an effective <strong>adaptive immune response.<\/strong><\/p>\n<h3><strong>Basic Structure of an Antibody<\/strong><\/h3>\n<p>All antibodies have a <strong>common structure<\/strong> consisting of two identical <strong>light chains<\/strong> and two heavy chains. Each chain consists of <strong>Ig domains<\/strong> (of approximately 110 amino acids) containing conserved sequences and <strong>intra-chain disulphide bonds.<\/strong> It is the V regions of the light and heavy chains that vary between antibodies and are responsible for<strong> antigen specificity.<\/strong><\/p>\n<h3><strong>Classification of Antibodies<\/strong><\/h3>\n<p>Antibodies are divided into several <strong>isotypes <\/strong>and <strong>subtypes<\/strong> according to differences in the <strong>C regions<\/strong> of the <strong>heavy chains<\/strong>. These isotypes include <strong>IgM, IgD, IgG, IgE and IgA<\/strong>, each with distinct functional properties. In addition, the two light chains of an antibody are of the same isotype (\u03ba or \u03bb), which are differentiated by their <strong>C domains.<\/strong><\/p>\n<h3><strong>Effector Function of Antibodies<\/strong><\/h3>\n<p>Most of the effector functions of antibodies are mediated through <strong>the C-regions<\/strong> of the <strong>heavy chains<\/strong>, although these functions are triggered by <strong>antigen binding<\/strong> to the binding site in the V-region. This binding can trigger a variety of <strong>immune responses<\/strong>, such as <strong>pathogen neutralisation<\/strong> and activation of the complement system.<\/p>\n<h3><strong>Monoclonal antibodies<\/strong><\/h3>\n<p><strong>Monoclonal antibodies<\/strong> are antibodies that are derived from a single <a href=\"https:\/\/www.cancer.gov\/espanol\/publicaciones\/diccionarios\/diccionario-cancer\/def\/linfocito-b\"><strong>B-lymphocyte clone <\/strong><\/a>and have specificity for a single <strong>antigenic determinant<\/strong>. These antibodies can be produced in the laboratory and are widely used in <strong>research, diagnosis and therapy<\/strong>. Their ability to recognise a single antigenic structure makes them powerful tools in <strong>modern medicine.<\/strong><\/p>\n<h3><strong>Antigen-Antibody Interaction<\/strong><\/h3>\n<p>Antigens are substances that specifically bind to <strong>antibodies or antigen receptors<\/strong> on <strong>T-lymphocytes<\/strong>. These antigens include a wide variety of biological molecules, such as sugars, lipids, proteins and nucleic acids. In contrast, <strong>T-lymphocyte receptors<\/strong> generally recognise only antigenic peptides.<\/p>\n<p><strong>Macromolecular antigens<\/strong> contain multiple <strong>epitopes <\/strong>or <strong>antigenic determinants<\/strong> that can be recognised by antibodies. <strong>Linear epitopes<\/strong> consist of adjacent amino acid sequences, whereas conformational determinants are formed by the folding of the <strong>polypeptide chain.<\/strong><\/p>\n<h3><strong>Affinity and Avidity in Antigen-Antibody Binding<\/strong><\/h3>\n<p>The <strong>affinity<\/strong> between an antibody and its epitope is usually represented by the <strong>Kd value.<\/strong> <strong>Polyvalent antigens<\/strong> have multiple identical epitopes to which several antibodies can bind. In the case of<strong> IgM, <\/strong>up to <strong>10 antibodies<\/strong> can bind simultaneously to the same <strong>antigen,<\/strong> which increases the avidity of the <strong>antibody-antigen interaction.<\/strong><\/p>\n<h3><strong>Changes in Antibody Structure During the Immune Response<\/strong><\/h3>\n<p>During an <strong>immune response,<\/strong> antibodies can undergo <strong>structural changes.<\/strong> Initially, <strong>B cells<\/strong> produce <strong>membrane-bound antibodies.<\/strong> However, as B cells are activated and become plasma cells, they begin to <strong>secrete antibodies<\/strong> with the same <strong>antigenic specificity<\/strong>. Changes in the <strong>C regions<\/strong> of antibodies without altering the <strong>V regions <\/strong>are the basis of isotype switching. This allows the effector function of the antibody to be modified without changing its <strong>specificity. <\/strong>In addition, point mutations in the V-regions lead to an increase in affinity for the antigen, a process known as <strong>affinity maturation.<\/strong><\/p>\n<h3><strong>Conclusion<\/strong><\/h3>\n<p>Antibodies are essential molecules for the body&#8217;s <strong>protection <\/strong>against <strong>pathogens,<\/strong> and their <strong>structural <\/strong>and <strong>functional diversity<\/strong> is key to the success of the adaptive immune system.
