{"id":39587,"date":"2026-04-09T13:40:35","date_gmt":"2026-04-09T11:40:35","guid":{"rendered":"https:\/\/immunostep.com\/?page_id=39587"},"modified":"2026-04-09T14:26:52","modified_gmt":"2026-04-09T12:26:52","slug":"complejo-cd3","status":"publish","type":"page","link":"https:\/\/immunostep.com\/es\/complejo-cd3\/","title":{"rendered":"COMPLEJO CD3"},"content":{"rendered":"<div>\n<h1>Detecci\u00f3n de la clonalidad de las c\u00e9lulas T:<\/h1>\n<\/div>\n<div>\n<p>La identificaci\u00f3n exhaustiva de los subconjuntos de c\u00e9lulas T permite realizar diagn\u00f3sticos de c\u00e1ncer m\u00e1s precisos.<\/p>\n<p>Para ello, ofrecemos reactivos de alta especificidad y sensibilidad que permiten realizar an\u00e1lisis eficaces y seguros mediante citometr\u00eda de flujo.<\/p>\n<\/p>\n<\/div>\n<p>\n    <a href=\"#findout\">Descubre c\u00f3mo funciona<\/a>\n<\/p>\n<div>\n<\/p>\n<p style=\"text-align: center;\">El complejo <b>CD3-TCR<\/b> interviene en el desarrollo y la activaci\u00f3n de los linfocitos T.<\/p>\n<p style=\"text-align: center;\">Est\u00e1 formado por la <b>mol\u00e9cula dim\u00e9rica TCR<\/b> (receptor de c\u00e9lulas T) y la <b>mol\u00e9cula hexam\u00e9rica CD3.<\/b><\/p>\n<p style=\"text-align: center;\"><b>CD3<\/b> est\u00e1 formado por cuatro cadenas diferentes, \u03b3, \u03b4, \u03b5 y \u03b6, que se combinan para formar 3 d\u00edmeros: un <b>homod\u00edmero \u03b6\/\u03b6<\/b> y <b>dos heterod\u00edmeros<\/b>, <b>\u03b3\/\u03b5 y \u03b4\/\u03b5.<\/b><\/p>\n<p style=\"text-align: center;\">Las cadenas \u03b3, \u03b4 y \u03b5 tienen una regi\u00f3n <b>ITAM<\/b> (motivo inmunoestimulador basado en tirosina), y la cadena \u03b6 tiene tres ITAM; esta regi\u00f3n interviene en la activaci\u00f3n celular. El complejo CD3 act\u00faa como correceptor del TCR cuando reconoce un ant\u00edgeno y participa en la se\u00f1alizaci\u00f3n interna de los linfocitos.<\/p>\n<p style=\"text-align: center;\">El TCR puede estar formado por una <b>cadena \u03b1<\/b> y una <b>cadena \u03b2 (TCR \u03b1\/\u03b2)<\/b> o por una cadena \u03b3 y una cadena \u03b4 (TCR \u03b3\/\u03b4), siendo la mayor\u00eda de los linfocitos T de la sangre perif\u00e9rica del tipo \u03b1\/\u03b2 y los del tipo \u03b3\/\u03b4 m\u00e1s abundantes en el tejido epitelial. Ambas cadenas tienen una regi\u00f3n constante (C) y una regi\u00f3n variable (V), correspondiendo esta \u00faltima al sitio de uni\u00f3n al ant\u00edgeno que recibe el p\u00e9ptido presentado por el <b>MHC (Complejo Mayor de Histocompatibilidad)<\/b> de las c\u00e9lulas presentadoras de ant\u00edgenos.<\/p>\n<p style=\"text-align: center;\">En el caso del <b>TCR \u03b1\/\u03b2,<\/b> la regi\u00f3n constante de la cadena \u03b2 presenta dos tipos, \u03b21 o \u03b22, por lo que se pueden distinguir dos poblaciones de <b>linfocitos T \u03b1\/\u03b2: TCR C\u03b21 (o TRBC1) y TCR C\u03b22 (o TRBC2).<\/b><\/p>\n<\/div>\n<div>\n<h1>Complejo CD3-TCR<\/h1>\n<\/div>\n<p><img decoding=\"async\" src=\"\/wp-content\/uploads\/2026\/04\/FINAL-FLYER-7.png\" alt=\"\"><\/p>\n<ul>\n<li>\n<h3>CD3\u03b4<\/h3>\n<div>\n<p>Adem\u00e1s de su papel en la activaci\u00f3n, interviene en el correcto <b>ensamblaje de los receptores de c\u00e9lulas T (TCR)<\/b> y en su expresi\u00f3n extracelular a trav\u00e9s de la <b>v\u00eda de activaci\u00f3n de la ERK (quinasa regulada por se\u00f1ales extracelulares)<\/b>, lo cual es importante para la selecci\u00f3n positiva de los timocitos.<\/p>\n<\/div>\n<\/li>\n<li>\n<h3>CD3\u03b5<\/h3>\n<div>\n<p>Desempe\u00f1a un papel en la regulaci\u00f3n de la <b>expresi\u00f3n del TCR<\/b> y en la selecci\u00f3n negativa t\u00edmica, y es un componente clave de la se\u00f1alizaci\u00f3n celular.<\/p>\n<\/div>\n<\/li>\n<li>\n<h3>CD3\u03b3<\/h3>\n<div>\n<p>Desempe\u00f1a un papel en la activaci\u00f3n celular, en el ensamblaje inicial del TCR y en la regulaci\u00f3n de su expresi\u00f3n en la superficie celular <b>a trav\u00e9s de la v\u00eda de activaci\u00f3n de la ERK (quinasa regulada por se\u00f1ales extracelulares)<\/b>.<\/p>\n<\/div>\n<\/li>\n<li>\n<h3>CD3\u03b6<\/h3>\n<div>\n<p><b>El d\u00edmero \u03b6\u03b6, CD247,<\/b> tiene tres ITAM por cadena y participa tanto en la activaci\u00f3n celular como en la diferenciaci\u00f3n intrat\u00edmica.<\/p>\n<\/div>\n<\/li>\n<li>\n<h3>ITAM (Immunoreceptor Tyrosine-based Activation Motif)<\/h3>\n<div>\n<p>V\u00edas de se\u00f1alizaci\u00f3n celular que, mediante la fosforilaci\u00f3n por parte de tirosina quinasas espec\u00edficas, permiten la <b>activaci\u00f3n celular<\/b>.<\/p>\n<\/div>\n<\/li>\n<li>\n<h3>CD3<\/h3>\n<div>\n<p>Una mol\u00e9cula esencial para el <b>ensamblaje y la regulaci\u00f3n del TCR<\/b> y para la <b>activaci\u00f3n de los linfocitos T<\/b>. Est\u00e1 formada por seis cadenas: \u03b3, \u03b4, \u03b5 (dos cadenas) y \u03b6 (dos cadenas).<\/p>\n<\/div>\n<\/li>\n<li>\n<h3>TCR (T Cell Receptor)<\/h3>\n<div>\n<p>Posee sitios de uni\u00f3n espec\u00edficos en su regi\u00f3n variable para los ant\u00edgenos presentados por las c\u00e9lulas a trav\u00e9s de la <b>interacci\u00f3n MHC-TCR,<\/b> lo que permite el reconocimiento espec\u00edfico y la activaci\u00f3n funcional de la c\u00e9lula para desencadenar una respuesta inmunitaria. Est\u00e1 compuesto por una <b>cadena \u03b1<\/b> y una <b>cadena \u03b2;<\/b> esta \u00faltima puede clasificarse como \u03b21 o \u03b22.<\/p>\n<\/div>\n<\/li>\n<\/ul>\n<hr>\n<div>\n<h5 class=\"uk-heading-bullet uk-text-center\" data-id=\"page#6-0-1-0\">CONTEXTO\/RELEVANCIA CL\u00cdNICA<\/h5>\n<\/div>\n<div>\n<p>El diagn\u00f3stico de las neoplasias de c\u00e9lulas T se ha beneficiado enormemente del desarrollo de anticuerpos dirigidos contra la regi\u00f3n constante de la cadena \u03b2 (C\u03b2) de la mol\u00e9cula del receptor de c\u00e9lulas T (TCR), lo que ha permitido analizar la distribuci\u00f3n de las poblaciones C<i>\u03b2<\/i>1 y C<i>\u03b22<\/i> e identificar anomal\u00edas que puedan indicar una enfermedad.<\/p>\n<\/div>\n<p><img decoding=\"async\" src=\"\/wp-content\/uploads\/2026\/04\/TCR-C%CE%B221.png\" alt=\"TCR C\u03b22\"><\/p>\n<div>\n<\/p>\n<p>Este m\u00e9todo de detecci\u00f3n es similar al modelo de diagn\u00f3stico de la neoplasia de c\u00e9lulas B, que se basa en la restricci\u00f3n de las cadenas ligeras de inmunoglobulina de membrana a las cadenas kappa (<i>\u03ba<\/i>) o lambda (<i>\u03bb<\/i>).<\/p>\n<p>El uso simult\u00e1neo de ambos anticuerpos (TCR C<i>\u03b2<\/i>1 y TCR C<i>\u03b2<\/i>2) garantiza la separaci\u00f3n correcta y espec\u00edfica de las poblaciones para identificar con precisi\u00f3n la relaci\u00f3n entre ambas.<\/p>\n<p>De este modo, es posible detectar si existe clonalidad, es decir, una distribuci\u00f3n completa o abrumadoramente predominante hacia un \u00fanico tipo de cadena <i>\u03b2<\/i>, lo que podr\u00eda ser indicativo de una neoplasia de c\u00e9lulas T.<\/p>\n<\/p>\n<\/div>\n<p>\n    <a href=\"https:\/\/immunostep.com\/es\/immunostep\/contacto\/\">CONTACTA CON NUESTROS EXPERTOS<\/a>\n<\/p>\n<div>\n<h5 class=\"uk-heading-bullet uk-text-center\" data-id=\"page#6-0-1-0\" style=\"text-align: center;\">AN\u00c1LISIS ESTRAT\u00c9GICO<\/h5>\n<\/div>\n<p><img decoding=\"async\" src=\"\/wp-content\/uploads\/2026\/04\/TCR-Landing.pptx.jpg\" alt=\"\"><\/p>\n<div>\n<h6 style=\"text-align: center;\">Ejemplo de una estrategia de an\u00e1lisis para un donante sano muestra de sangre perif\u00e9rica; se ha detectado clonalidad de las c\u00e9lulas T colaboradoras (CD4+) TCR C\u03b22).<\/h6>\n<p>En el caso de los linfocitos T, la poblaci\u00f3n espec\u00edfica se identifica mediante <b>anti-CD3<\/b> y las subpoblaciones se analizan seg\u00fan pertenezcan al <b>TCR C<i>\u03b2<\/i>1<\/b> o al <b>TCR C<i>\u03b2<\/i>2<\/b>. Es recomendable realizar un inmunofenotipado m\u00e1s preciso, separando primero las poblaciones de TCR <i>\u03b1\/\u03b2<\/i> y TCR <i>\u03b3\/\u03b4<\/i>, para identificar posteriormente las subpoblaciones de TCR <i>\u03b1\/\u03b2<\/i>: <b>CD4+\/CD8-<\/b> (c\u00e9lulas T colaboradoras o Th), <b>CD4-\/CD8+<\/b> (c\u00e9lulas T citot\u00f3xicas o Tc), <b>CD4-\/CD8-<\/b> (c\u00e9lulas T doblemente negativas o DN) y <b>CD4+\/CD8+<\/b> (c\u00e9lulas T doblemente positivas o DP).<\/p>\n<p><b>Los linfocitos T TCR <i>\u03b3\/\u03b4<\/i><\/b> identificados tambi\u00e9n pueden analizarse junto con los dem\u00e1s marcadores auxiliares.<\/p>\n<p>El uso de reactivos auxiliares permite un diagn\u00f3stico m\u00e1s fiable y preciso, ya que proporciona informaci\u00f3n detallada sobre todas las poblaciones posibles y su cuantificaci\u00f3n exacta. Es adecuado utilizar anticuerpos <b>TCR <i>\u03b1\/\u03b2<\/i>, TCR <i>\u03b3\/\u03b4<\/i>, CD4 y CD8<\/b> en combinaci\u00f3n con <b>TCR C<i>\u03b2<\/i>1 y C<i>\u03b2<\/i>2<\/b>, y otros marcadores de poblaci\u00f3n (tanto espec\u00edficos de c\u00e9lulas T como no espec\u00edficos) y marcadores de maduraci\u00f3n para detectar una expresi\u00f3n an\u00f3mala o ausente, lo que proporciona una procedimiento de lisis de los RBCs afectar al n\u00famero de leucocitos presentes en la muestra?<\/h3>\n<div>\n<p>No, el n\u00famero de leucocitos no se ve afectado y <strong>se recomienda lisar todas las muestras<\/strong>, de acuerdo con las <strong>directrices de EUROFLOW<\/strong>, incluyendo aquellas muestras de LCR en las que no haya contaminaci\u00f3n visible con RBCs. En este sentido, no se observan diferencias significativas entre el paso de lisis (FACS Lysing) y el paso de lavado con PBS, siendo la manipulaci\u00f3n de la muestra, y en particular la decantaci\u00f3n del sobrenadante, y no la lisis, la principal causa de p\u00e9rdida celular, por lo que se recomienda aspirar y no decantar los sobrenadantes tras los pasos de
